Working field:

Implementation of a 2-D HPLC system and a QTOF mass spectrometry in the environment of an automated bioprocess development lab.

Here we advertise two positions to complete our KIWI-biolab team with bioanalytical experts that are competent in the analysis of biomolecules by liquid chromatography and mass spectrometry.

The project aims for the smart integration of LC/MS into our high throughput bioprocess development facility (KIWI-biolab) and to develop standardized workflows and a shared database that would enable a holistic optimisation of bioprocesses across units and scales.
The focus is on a fully automated and properly integrated analysis workflow of LC/MS devices that offer:
(i) optimal performance for the respective scientific question,
(ii) High throughput solutions to solve the current bottlenecks with adequate LC/MS performance
criteria,
(ii) a well-structured and standardized strategy for data management and sharing according to FAIR data principles.

The feasibility and potential of the integrated use of LC/MS to follow the process dynamics in parallel bioreactors and to detect critical quality attributes (CQAs) is demonstrated at two exemplary levels:
(1) Metabolomic approach: Integrated HT analysis of medium components and metabolites adopting our current offline analytical LC/MS methods for
(i) short-chain carboxylic acids,
(ii) quantitative extracellular and intracellular amino acid analysis,
(iii) nucleotides, including cellular alarmons such as cyclic AMP and ppGpp.
(2) Proteomic approach:
(i) analysis of secreted and released host cell proteins (HCPs) in the cultivation medium,
(ii) proteome analysis for the (semi)quantitative analysis of marker proteins,
(iii) product- and process relevant interactome analysis (subunit interaction, inclusion body structure and composition).

The project will be conducted in close cooperation with other scientists in the KIWI-biolab which are specialists on automation, robotics, FAIR data management and mathematical modelling.

Among the main tasks are:
  • Integration of an 1290 Infinity II Bio 2D-LC / 6546 LC/MS QTOF system from Agilent into the KIWI-biolab environment including the implementation of the FAIR data management
  • Adaptation and development of the methods mentioned above for the analysis of fermentation media, amino acids, nucleotides and proteins
  • Implementation of the sample preparation and extraction methods on a liquid handling station
  • Practical advisor/supervison of Bachelor and Master theses

Requirements:

  • Successfully completed university degree (Master, Diploma or equivalent) in the field of biotechnology, chemistry or a related area
  • Proven experience in the area of bioanalytics and a focus on liquid chromatography and mass spectrometry advantage.
  • A finalised PhD degree is an advantage but not necessary
  • Independent scientific work in the area of analytical biotechnology or chemistry with a focus on the HPLC analysis and/or mass spectrometry of at least one of the following substance classes: sugars, amino acids, nucleosides/nucleotides, proteome
  • Experience in the analysis of pharmaceutically relevant critical protein product quality attributes (glycosylation, disulfide bind pattern, MS-based interaction based research) is an advantage
  • The candidate should be open-minded
  • The ability to teach in German and/or in English is required; willingness to acquire the respective missing language skills
  • The willingness to actively participate in scientific events is a pre-requisite
